Background: RNA interference has emerged as a new and potent tool to knockdown the expression of target genes and to investigate their functions. For short time experiments with mammalian cell lines, RNA interference is typically induced by transfecting small interfering RNAs (siRNAs). Primary cells constitute important experimental systems in many studies because of their similarity to their in vivo counterparts; however, transfection of these cells has been found to be difficult. Flow cytometric kinetic assay of the activity of Na+/H+ antiporter in mammalian cells.

Background The Na+/H+ exchanger (NHE) of mammalian cells is an integral membrane protein that extrudes H+ ion in exchange for extracellular Na+ and plays a crucial role in the regulation of intracellular pH (pHi). Thus, when pHi is lowered, NHE extrudes protons at a rate depending of pHi that can be expressed as pH units/s. Methods To abolish the activity of other cellular pH-restoring systems, cells were incubated in bicarbonate-free Dulbecco's modified Eagle's medium buffered with HEPES. Near Infrared Spectroscopy Detection and Quantification of Herbal Medicines Adulterated with Sibutramine.

There is an increasing demand for herbal medicines in weight loss treatment. Some synthetic chemicals, such as sibutramine (SB), have been detected as adulterants in herbal formulations. In this study, two strategies using near infrared (NIR) spectroscopy have been developed to evaluate potential adulteration of herbal medicines with SB: a qualitative screening approach and a quantitative methodology based on multivariate calibration. Methomyl analogues with increased biological activity towards F7T maize mitochondria

Abstract Methomyl analogues were synthesized by substituting alkyl moieties (C 2 -C 21 ) in the place of the carbamic methyl. They were assayed on mitochondria isolated from male sterile (F 7 T) and male fertile (F 7 N) maize. They had no action on F 7 N mitochondria. The heptadecyl (C 17 ) and heneicosanyl (C 21 ) derivatives had no conspicuous effect on F 7 T mitochondria. By contrast, the ethyl, propyl, butyl, nonyl, tridecyl (C 13 ) and pentadecyl (C 15 ) derivatives had the same type of activity as Methomyl on F 7 T mitochondria, namely stimulation of NADH oxidation and inhibition of malate oxidation. Using the fibre structure of paper to determine authenticity of the documents: analysis of transmitted light images of stamps and banknotes.

A novel method is presented for distinguishing postal stamp forgeries and counterfeit banknotes from genuine samples. The method is based on analyzing differences in paper fibre networks. The main tool is a curvelet-based algorithm for measuring overall fibre orientation distribution and quantifying anisotropy. Using a couple of more appropriate parameters makes it possible to distinguish forgeries from genuine originals as concentrated point clouds in two- or three-dimensional parameter space.

GHB related acids are useful in routine casework of suspected GHB intoxication cases

Abstract GHB related acids (3,4-dihydroxy butyric acid, 2,4-dihydroxy butyric acid and glycolic acid) are produced through oxidative GHB metabolism. These analytes could be potential biomarkers to ensure the diagnosis of a GHB intoxication and even prolong the detection window. Within this study, forensic routine cases were measured to consider the potential of additional gas chromatographic mass spectrometric analysis on these acids. 17 GHB positive real cases (10 serum samples and 7 urine samples) and 40 cases with suspicion of drugging in DFC cases and negative GHB results (21 serum samples and 19 urine samples) were evaluated. Near‐field optical addressing of single molecules in coplanar geometry: a theoretical study

Photonic transfer through elongated optical structures of submicrometre section microfabricated at the surface of dielectric or semiconductor samples can be enhanced by an appropriate structuring of the local refraction index. We show from computerized simulations that both the light localization and the spectroscopic properties of such structures can be used to selectively excite, in coplanar geometry, individuals molecules located in the near-field.
